Cryopreserved, uncultured primary human hepatocytes isolated from liver tissue. Our primary hepatocytes have been validated for CYP induction. Data on the fold induction of CYP1A2, CYP2B6 and CYP3A4 are generated for each lot of cells and is stated in the Certificate of Analysis for each order. They have post-thaw viability and plateability of >70%. We also offer purified hepatic stellate cells, intra-hepatic biliary epithelial cells and cryopreserved human Kupffer cells.
Assay-Ready Expanded (ARE) human hepatocytes
Assay-Ready Expanded (ARE) hepatocytes are cryopreserved human hepatocytes that are easy to plate and ready to use for a wide range of applications including hepatic metabolism, compound uptake, CYP induction/inhibition, hepatotoxicity, genotoxicity and 3D co-culture studies. See the Application Data here! ARE Hepatocytes express CYP enzymes, are metabolically functional, are polarized and can be infected by the Hepatitis C virus.
ARE Hepatocytes are primary human hepatocytes that have been expanded via activation of the Oncostatin M pathway. Large batch sizes of hepatocytes from the same donor are available on request, providing a reliable and consistent source of primary hepatocytes. ARE Hepatocytes should be used directly for endpoint assays and cannot be expanded further since de-differentiation may occur.
Our expanded primary hepatocytes have high viability and >90% plateability when using ARE Hepatocyte Thawing Medium & Maintenance Medium.
For genotoxicity applications, we have validated hepatocytes (ax3703) that can be used for micronucleus assays when grown in ARE Hepatocyte Genotoxicity Assay Medium. This medium permits hepatocyte cell division allowing micronucleus formation to be observed when hepatocytes are exposed to genotoxic compounds.
Human liver sinusoidal endothelial cells
Assay-Ready Expanded (ARE) liver sinusoidal endothelial cells are easy to plate, cryopreserved liver endothelial cells that have been previously expanded in vitro. Our liver sinusoidal endothelial cells express CD31+ and Mannose receptor and are validated for uptake of Low Density Lipoprotein (LDL). Large batch sizes of liver endothelial cells from the same donor are available on request, ensuring consistency from preliminary experiments to high-throughput screening. ARE liver sinusoidal endothelial cells can be passaged twice in our optimized liver sinusoidal endothelial cell culture medium. The medium contains recombinant growth factors tailored for promoting liver endothelial cell viability and growth.
Human hepatic organoids
Derived from stem cells, the organoids are self-renewing and proliferative, allowing further expansion to meet the needs of your study. Their physiological responses are retained from their tissue of origin which provides relative toxicological data. Applicable in multiple bioscience applications these organoids are a valuable alternative to hepatic cell lines and primary hepatocytes.
